摘要: 目的:探讨快速检测C57BL/ 6Job 小鼠基因型方法。C57BL/ 6Job 小鼠是一种ob 基因突变种,ob 基因的点突变导致小鼠遗传性肥胖。该品系广泛用于肥胖及有关疾病研究。方法与结果:本文用PCR 及限制性内切酶法快速,准确地检测出C57BL/ 6J 小鼠同窝仔鼠的三种基因型:野生型( + / + ) ,杂合体型(ob/ + ) 和突变型(ob/ ob) 。结论:该法只需很少的生物材料,即一小段小鼠尾巴,适用任何年龄的小鼠,对于该品系小鼠的育种、肥胖机理和肥胖预防的研究很有实用意义。

Abstract: Purpose and Methods : The ob gene point mutant in C57BL/ 6J ob mice cause genetic obesity in all life. To improve the speed and efficiency of ob genotyping , we developed a method with PCR and rest riction endonuclease by property primer. Results and Conclusion : Three genotype (ob/ ob , ob/ + , + / + ) of the mouse are rapidly , differetiated with this assay. Since only a small biopsy specimen ( tail end) is needed to genotype ,the ob mouse at any age is typable and can be used in subsequent breeding or research.
